Barkley was born in the Bronx, New York, and is the son of Tonya Johnson and boxer Alibay Barkley. In 2001, Barkley and his family moved to Bethlehem, Pennsylvania, in the Lehigh Valley. Later, the family migrated to Coplay, Pennsylvania, in 2005 after originally residing in nearby Allentown.

The state of Pennsylvania paid tribute to Barkley for his achievements as a player and his involvement in activities to help people around Pennsylvania. In 2018, the Pennsylvania legislature approved declaring March 14 as "Saquon Barkley Day" as a state holiday.

Whitehall High School, which Barkley attended, is a member of the Eastern Pennsylvania Conference. During his final year at Whitehall High School, Barkley ran for 1,856 yards and 31 touchdowns, and he rushed for 3,646 yards and 63 touchdowns. After being rated as a four-star prospect, Barkley committed to play football for Penn State. His first commitment was to Rutgers, but he changed to Penn State.

Barkley finished his freshman season with 1,076 rushing yards on 182 carries, averaging an impressive 5.9 yards per carry. Giants to a first playoff appearance in four years, starting all 16 of the games in the regular season. He finished the regular season with 1312 rushing yards, 10 rushing touchdowns, and a total of 295 rushing attempts.

On March 13, 2024, Barkley signed a three-year contract worth $37.75 million with the Philadelphia Eagles, which included a two-year guarantee of $26 million. With 15 touchdowns and a franchise-high 2,283 yards from scrimmage at the end of the season, Barkley led the league and contributed 34.8 percent of the team's overall output. He was chosen as a First-team All-Pro, won the Offensive Player of the Year title, and finished third in the MVP voting.
